重要提示:与其背景品系NOD/ShiLtJ(品系号001976)相似, NSG小鼠表现出进行性脱毛,到3月龄时通常已经很严重。

NOD.Cg-Prkdcscid Il2rgtm1Wjl/SzJ小鼠——通常以其商品名 NOD scid gamma (NSG) 为人所知——既不表达 Prkdc 基因,也不表达 X 连锁 Il2rg 基因。NSG 小鼠可存活且可繁育、体型正常,无任何可见的身体和行为异常。淋巴组织的组织学检查显示,胸腺中无淋巴样细胞和某些囊性结构,脾脏中无滤泡,淋巴结细胞结构显著减少。NSG 小鼠缺乏成熟淋巴细胞、无法检测到血清 Ig,自然杀伤 (NK) 细胞的细胞毒活性极低。该品系在经过亚致死辐射处理后也不易发生淋巴瘤。NSG 小鼠经证明可支持高效的人CD34+ 造血干细胞移植,适用于长期稳定的异种移植研究。NSG小鼠对于可致糖尿病的链佐霉素(STZ)高度敏感。

尾部扭曲,头部歪斜,转圈以及后肢无力在NSG品系中的发生率很低,在实验用未经处理的幼鼠中的发生率通常低于0.01%, 在经过人源化或其它研究处理的成年鼠中发生率通常低于0.5%。

这些双突变小鼠的培育方法如下:将雌性 NOD.CB17-Prkdcscid/J 小鼠(品系货号 001303)与携带 X 连锁 B6.129S4-Il2rgtm1Wjl/J 等位基因的雄性小鼠(品系货号 003174)杂交。将得到的 Prkdcscid 等位基因杂合子和 Il2rgtm1Wjl 等位基因半合子雄性小鼠与雌性 NOD.CB17-Prkdcscid/J 小鼠(品系货号 001303)杂交 8 代。将杂合子小鼠杂交产生 Prkdcscid 等位基因纯合子小鼠,Il2rgtm1Wjl 等位基因纯合子(雌性)或半合子(雄性)小鼠。
